Incidentally, it's also the only part of the song that actually calls for Petty to sing in something besides his usual talent-free breezy Dylan-esque manner.
As for the rest, well, whatever it was, it certainly wasn't pulled from the albums.
But as we learned with Journey, that fact alone shouldn't put anyone's doubts to bed.
That vocal delay is used every single time I've ever seen the band play that song.
Then it is most likely a sample, triggered by the keyboard player.
I was listening closely, and there were very audible artifacts from compressor / limiters, which smoothed out as the performance went along. By the last song, it was pretty good, for a live TV mix. I don't have a subwoofer or anything fancy, either. I did listen off the cable box on headphones for a bit though.
Robert Scovill, TP's engineer, pioneered the use of digital multi track tapes for sound checks, and I'm quite sure they know how to fill in the mix with sample, performers have been doing this for decades now.
